SEVENTH FRAMEWORK PROGRAMME

The Project - WP 07: Proteomic analysis of low dose radiation effects on the heart (HELMUC)

Objectives

The main goal of this Workpackage is the evaluation of the pathological changes in protein expression in cardiac endothelial cells after low radiation doses

Task 1:   Map the proteome changes in established and primary endothelial cells
             after low radiation doses (0.05Gy - 2Gy)
Task 2:   Validate key components of the radiation-specific changes in the proteome in the
             macro- and micro-vascular models of endothelial cell irradiation

Workpackage description

  • A series of established endothelial cell lines and primary endothelial cell cultures (WP2) will be irradiated in vitro at doses ranging from 2Gy to 50mGy. Afterwards proteins will be extracted and will be separated by 2D gel electrophoresis. Proteins showing radiation-specific changes will be excised and identified by MALDI -TOF mass spectrometry.
  • These key proteins will be validated by the identical technology using extracts from in vivo irradiated tissue samples from WP2 and the time course and dose response of changes in the radiation response markers of the endothelial cell proteome will be followed in vivo in the animal models of heart irradiation.

WP leader

Dr. Soile Tapio - leader of the Radiation Proteomics research group at the Helmholtz Centre Munich, Institute of Radiation Biology - is a radiation biologist specializing in protein purification and identification using 2D gel electrophoresis and MALDI-TOF mass spectrometry. The GSF has recently established a core proteome analysis facility (PAC) that will be available to the CARDIORISK project.
